ABSTRACT

ABSTRACT Lutzomyia intermedia (Diptera: Psychodidae) features as one of the main vectors that are involved in the transmission of American cutaneous leishmaniasis (ACL) in the Neotropical region. However, genetic studies involving this taxon are still incipient and important for understanding the level of variability of different populations, their role, and implications as vectors. The aim of this study was to determine the level of genetic diversity of L. intermedia present in the Ribeira River Valley, an area of ACL transmission in the state of Paraná, Brazil, through the Random amplified polymorphic DNA (RAPD). Two municipalities were chosen to collect sand flies: Cerro Azul (new transmission area of the ACL) and Adrianópolis (endemic area of the ACL). The insects were captured in the house, in the peridomicile and in the wild (forest). Two of the used markers made it possible to estimate the polymorphism of the studied populations, resulting in 40 genotypes, most of them from peridomicile. The dendrogram generated by the analysis with the primer A10 showed different degrees of similarity, suggesting that there may be gene flow in the studied populations. The Principal Coordinate Analysis (PCO) with the A2 primer, was useful in grouping L. intermedia according to its ecological and geographical origin. There was no distinction between the lineages composing the L. intermedia complex. The results of this study, with the record of great genotypic diversity in L. intermedia, may contribute to explain the maintenance of the life cycle of Leishmania braziliensis (Kinetoplastida: Trypanosomatidae) in the region.


RESUMEN Lutzomyia intermedia (Diptera: Psychodidae) es uno de los principales vectores que participan en la transmisión de leishmaniasis cutánea americana (LCA) en la región Neotropical. A pesar de que aún los estudios genéticos que involucran a este taxón son incipientes, tienen una gran importancia para comprender el nivel de variabilidad de las diferentes poblaciones y sus implicaciones en su papel vectorial. El objetivo de este estudio fue determinar el nivel de diversidad genética de L. intermedia presente en el Valle del Río Ribeira, área de transmisión de LCA en el estado de Paraná, Brasil, mediante RAPD (ADN polimórfico amplificado aleatoriamente). Los flebótomos fueron recolectados en los municipios Cerro Azul (nueva área de transmisión de LCA) y Adrianópolis (área endémica de LCA), donde fueron capturados en ambientes residenciales, en el peridomicilio y en el bosque. Dos de los marcadores utilizados permitieron estimar el polimorfismo en las poblaciones estudiadas con la obtención de 40 genotipos, la mayoría de ellos en el peridomicilio. El dendrograma generado por el análisis con el cebador A10 mostró diferentes grados de similitud, lo que sugiere que puede haber flujo gènico en las poblaciones. El Análisis de Coordenadas Principales (PCO) con el cebador A2 fue útil para agrupar L. intermedia según su origen ecológico y geográfico. No hubo distinción entre los linajes que componen el complejo L. intermedia. Los resultados de este estudio, con el registro de gran diversidad genotipica en L. intermedia, pueden contribuir a explicar el mantenimiento del ciclo biológico de Leishmania braziliensis (Kinetoplastida: Trypanosomatidae) en la región.

ABSTRACT

A new species of Moenkhausia is described from the rio Machado drainage, Amazon basin, Brazil. It is diagnosed from congeners by its color pattern, consisting of the concentration of chromatophores on the anterior portion of body scales, the horizontally elongate blotch on caudal peduncle, a bright golden coloration of the dorsal portion of eye when alive, and a dark line crossing the eye horizontally. The new species has variable morphology regarding trunk lateral-line canals. Most fully grown individuals do not have enclosed bony tube in many lateral line scales, resembling early developmental stages of tube formation of other species. This paedomorphic condition is interpreted as a result of developmental truncation. Such evolutionary process may have been responsible for the presence of distinct levels of trunk lateral line reductions in small characids. Variation in this feature is common, even between the sides of the same individual. We reassert that the degree of trunk lateral-line tube development must be used with care in taxonomic and phylogenetic studies, because reductions in the laterosensory system may constitute parallel loss in the Characidae. We suggest the new species to be categorized Near Threatened due to the restricted geographical distribution and continuing decline in habitat quality.(AU)


Uma espécie nova de Moenkhausia é descrita da drenagem do rio Machado, bacia Amazônica, Brasil. É diagnosticada das congêneres pelo padrão de coloração, que consiste na concentração de cromatóforos na porção anterior das escamas do corpo, em uma mancha horizontalmente alongada no pedúnculo caudal, na coloração dourada brilhante da porção dorsal do olho quando vivo e na faixa escura que atravessa o olho horizontalmente. A nova espécie apresenta variação na morfologia do canal da linha lateral do corpo. A maioria dos indivíduos totalmente desenvolvidos não possuem tubo ósseo fechado em muitas escamas da linha lateral, assemelhando-se aos estágios iniciais do desenvolvimento da formação do tubo de outras espécies. Essa condição pedomórfica é interpretada como resultado do truncamento do desenvolvimento. Tal processo evolutivo pode ter sido responsável pelos diferentes níveis de redução do canal sensorial de pequenos caracídeos. A variação neste caráter é comum, até entre os lados do mesmo indivíduo. Por isso, reafirmamos que o grau de desenvolvimento do canal sensorial do corpo deve ser usado com cuidado em estudos taxonômicos e filogenéticos, porque reduções no sistema látero-sensorial podem significar perdas paralelas em Characidae. Sugerimos que a espécie nova seja categorizada como Quase Ameaçada devido à distribuição geográfica restrita e ao declínio contínuo da qualidade do habitat.(AU)

ABSTRACT

The ripe dried fruit of citron(Citrus medica) is one of the important sources of Chinese herb Citri Fructus. At the same time, it is also grown for edible and ornamental uses. There are many species and abundant genetic variation. To clarify the intraspecific variation and resource distribution of citron, this study investigated the variation in 11 citron fruits, basically covering the main species in China, including Xiaoguo citron(C. medica var. ethrog), Goucheng(C. medica var. yunnanensis), Muli citron(C.medica var. muliensis), Dehong citron(C.medica×Citrus spp.), Fuzhou citron(C.medica×C.grandis?), Mawu(C.medica×C.grandis?), Cangyuan citron, Binchuan citron, Sweet citron, Big citron, and Small citron. The natural communities of citron were proved to be mainly distributed in the southwestern and western Yunnan and southeastern Tibet of China, with Yunnan, Sichuan, Guangxi, Chongqing, Hubei, and Zhejiang identified as the main production areas. Citron has also been widely grown in India, the Mediterranean region, and the Caribbean coast countries. The field investigation revealed the large-scale intraspecific variation of citron fruits. Most of the fruits are oval-like or sphere-like in shape. The fruits are green when raw and yellow when ripe, with oil cell dots on the skin, stripe-likes running from top to bottom, and bulge at the top. Usually, in the smaller citron fruits, the pulp and juice vesicles are better developed and the central columella is tighter. By contrast, the juice vesicles and central columella in larger fruits became more vacant, with carpels visible, and the apex segregation and development of the carpels is one of the reasons for variation. These variations should be given top priority in the future variety selection and breeding, and the quality differences of different citron species and their mechanisms should be further studied. In particular, variety selection and classification management according to their medicinal or edible purposes will provide scientific and technological supports for the orderly, safe, and effective production of citron products consumed as food and medicine.

ABSTRACT

Hyphessobrycon itaparicensis was originally described from a small stream in the Itaparica coastal island, Bahia State, Brazil, and has been sampled in several coastal rivers draining Bahia and Sergipe States. Broad examination of type material and recently collected specimens resulted in the redescription provided herein. The presence of one humeral blotch, absence of pseudotympanum, and conservation aspects of H. itaparicensis are briefly discussed. Data obtained from the original description, paratypes and topotypes of H. sergipanus showed broad overlap with H. itaparicensis and absence of morphological diagnostic features supporting the recognition of H. sergipanus as a valid species. Thus, H. sergipanus is considered as junior synonym of H. itaparicensis. We emphasize the need of examining large population samples and type material of similar congeners to avoid improper propositions of new specific names.(AU)


Hyphessobrycon itaparicensis foi descrita de um pequeno riacho na Ilha de Itaparica, Estado da Bahia, Brasil, e tem sido amostrada em outros rios costeiros nos estados da Bahia e Sergipe. Um amplo exame do material tipo e exemplares recentemente coletados resultou na redescrição aqui apresentada. A presença de uma mancha umeral, a ausência de pseudotímpano e aspectos da conservação de H. itaparicensis são brevemente discutidos. Dados obtidos da descrição original, de parátipos e topótipos de H. sergipanus revelaram ampla sobreposição com H. itaparicensis e ausência de características morfológicas diagnósticas que sustentem o reconhecimento de H. sergipanus como espécie válida. Assim, H. sergipanus é considerada sinônimo júnior de H. itaparicensis. Nós enfatizamos a necessidade de examinar grandes amostras das populações e do material tipo de congêneres semelhantes para evitar a proposição indevida de nomes novos de espécies.(AU)

ABSTRACT

Based on the form-function interaction and its consequence to niche exploitation by fish species, the study aimed to identify ecomorphological patterns and to investigate the possibility of explaining the trophic niche breadth using the pattern of intraspecific ecomorphological diversity. We tested the following hypotheses: i) the morphology explains variations in diet among fish species; ii) the intraspecific ecomorphological diversity is related to the breadth of the trophic niche explored by the species, so that species that feed on a wider range of resources have greater variation in body shape compared to those specialized in resource consumption. Fish were collected in stands of the aquatic macrophytes Eichhornia azurea and Eichhornia crassipes in lentic environments of the Upper Paraná River floodplain, Brazil. Two major trends were observed in the morphological space: fish with body shapes adapted to explore the substrate and others with a design that facilitates the capture of food items in more structured habitats. The relationship of diet with body shape was confirmed by significant relationships between matrices of trophic and morphological distances, providing evidence that morphology is related to interspecific variations in the use of trophic resources. However, the ranges of morphological and intraspecific trophic variations were not significantly related, rejecting the second hypothesis about the relationship between intraspecific ecomorphological diversity and trophic niche breadth. The morphological characteristics often have multiple ecological roles, which could result in trade-off among these functions. Thus, fish with highly specialized morphology may show specialist feeding or even generalist habit, because in this case some resources may be difficult to exploit, even by a specialist. Species with low and high morphological diversity demonstrated narrow trophic niche and the availability of resources may have been essential for consumption. Therefore, species morphology is related to the use of food resources and ecomorphology can be considered an important tool for the prediction of the exploited niche space by species in assemblages. However it is not possible to predict if species with greater intraspecific morphological diversification indeed have wider niche, since the abundance or scarcity of the available food resources may interfere with trophic niche breadth.(AU)


Baseado na interação entre forma e função e suas consequências para exploração do nicho pelas espécies, o objetivo deste estudo foi identificar padrões de variação ecomorfológica e investigar se a amplitude de nicho trófico pode ser explicada pelo padrão de diversificação ecomorfológica intraespecífica. Assim, testaram-se as hipóteses: i) a morfologia explica as variações na dieta entre as espécies de peixes ii) a diversificação morfológica intraespecífica está relacionada com a amplitude do nicho trófico explorado pelas espécies, de modo que, espécies que se alimentam de maior variedade de recursos apresentam maior variação na forma do corpo em relação aquelas especializadas no consumo de recursos. Os peixes foram coletados em bancos de macrófitas aquáticas de Eichhornia azurea e Eichhornia crassipes em ambientes lênticos da planície alagável do alto rio Paraná, Brasil. Duas tendências principais foram observadas no espaço morfológico: peixes com formato corporal adaptado a explorar o substrato e outros com forma do corpo facilitando a captação de alimentos em hábitats mais estruturados. A relação da dieta com a forma do corpo foi confirmada por relações significativas entre as matrizes de distâncias trófica e morfológica, evidenciando que a morfologia pode estar relacionada às variações interespecíficas no uso de recursos alimentares. No entanto, as amplitudes de variações morfológicas e tróficas intraespecíficas não estiveram significativamente relacionadas, rejeitando-se a segunda hipótese sobre a relação entre diversidade ecomorfológica intraespecífica e amplitude do nicho trófico. Características morfológicas podem expressar diferentes papéis ecológicos, o que poderia resultar em troca entre estas funções. Assim, um peixe com morfologia altamente especializada pode ter alimentação especialista ou até mesmo hábito generalista, porque, neste caso, alguns recursos podem ser difíceis de explorar, mesmo por um especialista. Espécies com pequena e grande diversidade morfológica apresentaram nicho trófico estreito e a disponibilidade dos recursos pode ter sido decisiva para o consumo. Portanto, a morfologia das espécies está relacionada com o uso de recursos alimentares, de modo que a ecomorfologia pode ser considerada uma ferramenta importante na predição da ocupação do espaço de nicho pelas espécies em assembleias. No entanto, não é possível prever se espécies que apresentam maior diversidade de formas entre seus indivíduos (i .e ., maior diversificação morfológica intraespecífica), de fato apresentam nichos mais amplos, uma vez que a abundância ou escassez do recurso alimentar disponível no ambiente pode interferir na amplitude do nicho trófico ocupado.(AU)

ABSTRACT

We tested whether interindividual variations in diet composition within a population of Deuterodon stigmaturus can be explained by morphological differences between individuals, and whether diet-morphology relationships are dependent on the ontogenetic development. We analyzed diet of 75 specimens sampled in a coastal stream of Southern Brazil. Variation in stomach content was summarized with a Principal Coordinate Analysis (PCoA). The retained PCoA axes were tested as response to standard length (SL), and to values of intestine length (IL) and mouth length (ML) independent of body size, using linear mixed-effects models (LMM). The most consumed food items by D. stigmaturus were filamentous algae (41%), terrestrial plants (20.3%), detritus (12%), and aquatic invertebrates (8.8%). The LMMs showed that SL was positively related to consumption of terrestrial plants, whereas IL independent of SL was negatively related to aquatic invertebrates and positively related to filamentous algae. When body sized was held constant, ML was not related to diet variation. Interindividual diet differences conditioned to body size suggest that individuals shift their trophic niche and function in the ecosystem along the ontogenetic development. Relationships between intestine length and diet composition suggest interindividual differences in foraging ability and digestibility of distinct food items.


Nós testamos se variações na composição da dieta de uma população de Deuterodon stigmaturus podem ser explicadas por diferenças morfológicas entre indivíduos, e se as relações entre morfologia e dieta são dependentes do desenvolvimento ontogenético. A dieta de 75 indivíduos amostrados em um riacho costeiro do sul do Brasil foi analisada. Uma Análise de Coordenadas Principais (PCoA) foi utilizada para sumarizar variações individuais no conteúdo estomacal. Os eixos retidos da PCoA foram testados como resposta ao comprimento padrão (CP) e ao comprimentos do intestino (CI) e da boca (CB), independentes do tamanho corporal, usando modelos lineares mistos (LMMs). Os itens macroscópicos mais consumidos por D. stigmaturus foram algas filamentosas (41%), plantas terrestres (20,3%), detritos (12%), e invertebrados terrestres (8,8%). Os LMMs mostraram que o CP foi positivamente relacionado ao consumo de plantas terrestres, enquanto que valores de CI independentes de CP foram negativamente relacionados ao consumo de invertebrados aquáticos e positivamente relacionados ao consumo de algas filamentosas. Quando o CP foi mantido constante, CB não foi relacionado a composição da dieta. As diferenças na dieta condicionadas ao tamanho corporal sugerem que os indivíduos mudam seu nicho trófico e função no ecossistema ao longo do desenvolvimento ontogenético. As relações entre o tamanho do intestino e composição da dieta sugerem diferenças individuais na habilidade de forragear e digerir itens alimentares distintos.

ABSTRACT

Variation in body characteristics related to lizard locomotion has been poorly studied at the intraspecific level in Anolis species. Local adaptation due to habitat heterogeneity has been reported in some island species. However, studies of mainland species are particularly scarce and suggest different patterns: high variability among highland lizards and poorly differentiated populations in one Amazonian species. We characterized inter population variation of body size and shape in the highland Andean Anolis ventrimaculatus, an endemic species from Western Colombia. A total of 15 morphometric variables were measured in specimens from the reptile collection of the Instituto de Ciencias Naturales, Universidad Nacional, Colombia. The study included individuals from seven different highland localities. We found size and shape sexual dimorphism, both of which varied among localities. Patterns of variation in body proportions among populations were different in both males and females, suggesting that either sexual or natural selective factors are different in each locality and between sexes. Since this species exhibits a fragmented distribution in highlands, genetic divergence may also be a causal factor of the observed variation. Ecological, behavioral, additional morphological as well as phylogenetic data, may help to understand the evolutionary processes behind the geographic patterns found in this species.


La diversificación fenotípica al interior de una especie en características de dimensiones corporales relacionadas con la locomoción de los lagartos, se ha estudiado poco en especies de Anolis. Los datos de algunas especies de isla revelan patrones distintos de variación geográfica y sugieren que la adaptación local, debida a la heterogeneidad del hábitat, ocurre a este nivel. Los estudios de especies de continente son particularmente escasos y sugieren patrones distintos: un lagarto altoandino altamente variable y poblaciones poco diferenciadas en una especie amazónica. Caracterizamos la variación inter poblacional en el tamaño y forma del cuerpo del lagarto altoandino Anolis ventrimaculatus, especie endémica del Oeste de Colombia. Encontramos variación geográfica en el dimorfismo sexual en tamaño y forma. El patrón de variación en las proporciones corporales entre poblaciones fue distinto en machos y en hembras, sugiriendo que las presiones de selección sexual o natural son diferentes en cada localidad. Dado que la especie exhibe una distribución fragmentada en alta montaña, la divergencia genética entre poblaciones puede ser otro factor causal de la variación observada. Datos ecológicos, etológicos y morfológicos adicionales, así como información filogenética puede contribuir al entendimiento de los procesos evolutivos responsables del patrón de variación geográfica encontrado en esta especie.

ABSTRACT

Enzyme activities of Cenococcum geophilum isolates were examined on enzyme-specific solid media. Deoxyribonuclease, phosphatase, and urease were detected in all isolates, whereas cellulase was not detected in any of the isolates. Variations in enzyme activities of amylase, caseinolysis, gelatinase, lipase, and ribonuclease were observed among isolates.

ABSTRACT

Characterization of all chromosomes of the Andean G19833 bean genotype was carried out by fluorescent in situ hybridization. Eleven single-copy genomic sequences, one for each chromosome, two BACs containing subtelomeric and pericentromeric repeats and the 5S and 45S ribosomal DNA (rDNA) were used as probes. Comparison to the Mesoamerican accession BAT93 showed little divergence, except for additional 45S rDNA sites in four chromosome pairs. Altogether, the results indicated a relative karyotypic stability during the evolution of the Andean and Mesoamerican gene pools of P. vulgaris.

ABSTRACT

Rhodnius pallescens is the main vector of Trypanosoma cruzi in Panama and one of the most relevant secondary vectors in Colombia. Despite the importance of this species, there is limited knowledge about the genetic variability along its geographical distribution. In order to evaluate the degree of karyotype variability we analyzed the meiotic behavior and banding pattern of the chromosomes of 112 males of R. pallescens coming from different regions of Colombia and Panama. Using the C-banding technique we identified two chromosomal patterns or cytotypes characterized by differences in the amount, size and distribution of constitutive heterochromatic regions in the chromosome complement (2n = 20 autosomes plus XY in males). The individuals can be easily classified in each cytotype by the analysis of the chromosomes during first meiotic prophase. The frequencies of the cytotypes are variable according to the geographic origin of the populations. This chromosomal divergence together with morphological data supports the existence of three genetically different populations of R. pallescens and provides new information to understand the distribution dynamics of this species.

Based on the results of comparative analyses of 1,039 specimens of several progenies of Anopheles nuneztovarifrom three localities in Colombia, eight costal wing spot patterns were observed. Patterns I and III were the most frequent: 77.96 percent and 11.36 percent, respectively. Using the diagnostic characters ratio of the length of the basal dark area of hind tarsomere II/length of hind tarsomere II, ratio of the length of the humeral pale spot/length of the pre-humeral dark spot, and the ratio of the length of the subcostal pale spot/length of the distal sector dark spot (DS-III2/Ta-III2, HP/PHD, SCP/DSD) approximately 5 percent of the adult females were misidentified as a species of Nyssorhynchus, different from An. nuneztovari. Approximately 5 percent of the specimens showed DS-III2/Ta-III2 ratio less than 0.25 (range 0.21 - 0.24), and among them 3.34 percent shared a HP/PHD ratio less than 1.50. Consequently, 1.52 percent of An. nuneztovari individuals can be misidentified as Anopheles oswaldoi. In those specimens with the DS-III2/Ta-III2 ratios higher than 0.25, 34.45 percent displayed SCP/DSD values greater than 0.50 and of these, 3.65 percent displayed HP/PHD values greater than 1.8. This combination of characters could lead one to misidentify samples of An. nuneztovari as Anopheles rangeli. Similarly, 2.43 percent of the females could be identified erroneously as either Anopheles aquasalis or Anopheles benarrochi. Individuals with a HP/PHD ratio greater than 2.0, could be misidentified as Anopheles trinkae, Anopheles strodei or Anopheles evansae. A distinct combination of diagnostic characters for An. nuneztovari from Colombia is proposed.
